Hi,

I want to add some samples to the SD. Do I have to make sure to turn off the nerdseq before remove SD ?

Removing is no problem at all. Inserting would work or not. Sometimes on the 2nd try loading.
Generally hot swapping of the sdcard is not supported but you can try it. 
Word of advice, backup your cart each time it is in the computer to be sure your projects are backed up.
