In general or in mapping?

I've the samples loaded beforehand.

Doesn’t matter how, via mapping, playing a sample once (which makes it the last used one. Loading samples is good but did you select the things to be used on slot 2? It doesn’t know which sample you want to trigger on which pitch and volume. It should work if you set that up once.
